How to use wp-categories as subscriber group

You cannot directly use WP categories as groups.

You would need to create different groups with name same as WP categories. Then you can set up notification on them by following steps from here – How to Configure and Send Notification Emails to subscribers when new posts are published

I have 500 users which are interested in post notification and 20 wp-categories. Does it imply that I have to manage 10.000 subscribers?
Can the subscribers unselect a group themselfes? How?

Is it right, that for each subscriber group I have to create a separate Post Notification (Email subject)? In consequence for 20 wp-categories I must have 20 notifications???

Subscribers cannot unselect a group themselves.
