Localhost to Remote host Permalink Setup Question

Hi, I am an amateur WordPress developer and I develop my own sites using WordPress. I am using WAMP for developing sites. Well most recently I have started using WP 4 version. Well now here is my problem…

I browse my website using localhost/WP_folder_name_in_www folder. But after installation of wordpress for a new site once I went to dashboard>settings>permalink and then checked “Post name” radio button my site actually crashed! It is happening when I am changing permalink to any other option every time. Actually why it is happening and how to fix the problem?

My second question is after development of a website on localhost once I upload the site in remote host I face extreme problem with link setup. Everytime I had to change permalinks manually! Is there any why which will take care of all permalinks of my site even if I transfer them from localhost to remote host? This broken link problem is actually very annoying one for me.
