Modify default post query?

Hello,

I was hoping someone can point me into the right direction to modify the default post query for a custom post type, in a plugin, to do the following;

1. Sort the posts by a custom meta data (the value is a number) attached to the posts.
2. Do the sorting through ajax, so it bypasses caching.
