Permalinks problem

After I set my permalink to Day and time, it pops up a message on the top of the screen.
“Permalink structure updated. Remove write access on web.config file now!”
“By default WordPress uses web URLs which have question marks and lots of numbers in them; however, WordPress offers you the ability to create a custom URL structure for your permalinks and archives. This can improve the aesthetics, usability, and forward-compatibility of your links. A number of tags are available, and here are some examples to get you started.”
Then after I reloaded the page, it returns an error page of “500 Internal Server error” “There is a problem with the resource you are looking for, and it cannot be displayed.”
Does anyone know what happen and can you tell me why?
