Permissions and editing files – gurus please respond

I have gone in and changed the permissions on my wordpress files, but wordpress will not acknowledge that the permissions have been changed. These are on blogs that have been constantly active for years and I have never had this problem with them before.

Please say there is someone with an answer for this.
