    Try flushing your permalink rewrite rules by visiting Settings > Permalinks and resaving the settings on that page

    Turns out that this was a symptom not a cause. Deactivating the plugin was removing the problem so it appeared that the plugin was causing the problem – not so.

    I’m also encountering the same problem. Flushing the permalinks does not help the issue. Only deactivating the plugin returns the site to normal operation.

    I just discovered this same problem this evening. i am using latest WordPress, with 0.7.2 plugin. I had to set default permalinks, deactivate all plugins, and delete htaccess.

    I set permalink back to what I want, then one by one activated the plugins. Once I turned on Custom Post Type UI the permalinks stopped working again (strangely only on the fist level, nested permalinks still worked), and I have to repeat the whole process to get them back (wouldn’t work by just turning to default, then back to custom).

    Any ideas? I had read a tidbit that maybe a flush had been removed from the plugin in 0.5.2?
