Using an image not in media library

Is it possible to create the colorbox effect when using an image that isn’t in my media library (but in a directory within my WP instance)?
If so, can you give me a foo code example of what the link tag would look like?
If not, I installed the plugin tried using an image in my media library as the image on the page itself but I didn’t see any field called “Link URL” as stated in your instructions: “When inserting a picture, the field “Link URL” needs to contain the link to the full-sized image. (press the button “Link to Image” below the field)”
So, I’m a bit confused on how to use this.
