It’s very difficult to tell you what options you should enable, as that depends on your personal requirements.
Before enabling one option, the best for you would be to read the help documentation for it first. All the documentation can be found here:

In regards to headers, we suggest you to read the following:

Hi,
You do not need to put in your WordPress URL. The value you put should be only the last part of the URL path.
Only letters and numbers are permitted, i.e. abc123def45. If you use any other characters, they will be filtered out.

– some other security plugin running on that site
– other plugin that modifies your login screen
– you have added your IP address to the white listHope that helps.

I’m not sure if you’re asking about WPMU or an option to have the same Shield settings on multiple sites automatically.
If your question was “Can Shield be used in Multisite WordPress (WPMU)?”, then the answer is yes, it can be network activated, but there are no options for subsites.
If your question was “Is there an option to setup the Shield Security plugin on 1 site and have all options replicated to other sites automatically?”, then the answer is yes. There is an option available (“Export/Import” Pro feature). Please see here:

Hello,
Email you received is for “Core Files Scanner”.
You can repair unrecognised WordPress core files automatically – go to Hack Protection module => Core File Scanner (enabled) => enable “Auto Repair” to set the scanner to automatically repair files when the scan next runs.
